Beschreibung

[N-Acetyl-Ser1]-Teriparatide is the N-terminally acetylated peptide impurity of teriparatide arising from modification of the Ser1 residue. The acetyl addition (+42.01 Da) neutralizes the N-terminal amine | alters peptide pI and hydrophobicity | and typically produces a reproducible retention time shift in reversed-phase HPLC. Formation can occur as a process-related or storage-related side reaction via residual acetyl donors or acylation chemistry during synthesis and workup. Analytical identification is achieved by LC-MS and MS/MS peptide mapping to confirm site-specific modification. Control measures include reagent specification | optimized deprotection protocols | scavengers and validated in-process and release assays to ensure impurity stays within specification.
