L-Pyroglutamylalanin, auch als Pidotimod-Verunreinigung 4 (Pyr-Ala-OH) bezeichnet, ist eine definierte prozessbezogene dipeptidische Verunreinigung, die dem cyclisierten N-terminalen Pyroglutamyl-Derivat von Alanin entspricht. Die Verbindung ist unter CAS 21282-08-6 erfasst und besitzt die Summenformel C8H12N2O4 bei einer Molekülmasse von 200,20 g/mol. Strukturell handelt es sich um ein synthetisches Nebenprodukt oder eine sequenzbezogene Verunreinigung, die während der Peptidkupplung oder durch Cyclisierung von Glutaminyl- beziehungsweise Glutamyl-Vorstufen entstehen kann. Aufgrund der hohen Polarität und fehlenden Flüchtigkeit sind für die Spurenbestimmung typischerweise LC-MS/MS sowie zur Bestätigung LC-UV oder hochauflösende MS erforderlich. Es handelt sich nicht um ein Nitrosamin, bleibt jedoch im Rahmen ICH-konformer Verunreinigungsprofile relevant.
