El L-piroglutamil alanina, también designado Impureza 4 de Pidotimod (Pyr-Ala-OH), es una impureza dipeptídica relacionada con el proceso y corresponde al derivado piriglutamilado cíclico en el extremo N-terminal de alanina. Se identifica con CAS 21282-08-6, fórmula molecular C8H12N2O4 y peso molecular de 200.20 g/mol. Su estructura es compatible con un subproducto de síntesis o una impureza relacionada con la secuencia, originada durante el ensamblaje peptídico o por ciclación de precursores glutaminilo o glutamilo. Por su carácter polar y no volátil, su cuantificación a nivel traza suele requerir LC-MS/MS, complementada con LC-UV o MS de alta resolución para confirmación ortogonal. No es una nitrosamina, aunque sigue siendo relevante en el perfil de impurezas bajo criterios reguladores ICH.
