Olodaterol is a long-acting β2-adrenergic receptor agonist (LABA) used for the maintenance treatment of chronic obstructive pulmonary disease (COPD). It provides bronchodilation for 24 hours by relaxing airway smooth muscles, improving airflow and reducing breathing difficulties. It is not intended for acute symptom relief but is used regularly to control chronic symptoms.
Olodaterol is administered via inhalation (e.g., Respimat® inhaler).
Indications include:
• Maintenance bronchodilator treatment of airflow obstruction in COPD, including chronic bronchitis and emphysema
• Not approved for asthma monotherapy
Parent: Olodaterol
Impurities in olodaterol arise from its multi-step chemical synthesis, involving aryl substitution, stereoselective synthesis, and salt formation, as well as from degradation due to light, oxidation, or hydrolysis. Due to its inhaled route and chronic use, impurity levels must strictly comply with ICH Q3A/B, USP, EP, and inhalation-specific regulatory guidelines.
Process-Related Impurities
• Residual intermediates such as aromatic aldehydes or substituted anilines
• By-products from chiral synthesis or amide bond formation
• Residual solvents including ethanol, acetonitrile, or toluene
Degradation Impurities
• Oxidation products of the phenolic or aromatic moieties
• Hydrolysis of the ether or amide linkages under high humidity or heat
• Photodegradation products, especially in aqueous or light-exposed formulations
• Stereoisomeric impurities due to racemization
Elemental Impurities
• Trace metals such as palladium, copper, or iron from catalysts or manufacturing equipment
• Controlled under ICH Q3D, with specific limits for inhalation exposure
Analytical Techniques for Impurity Detection
• High-Performance Liquid Chromatography (HPLC) – primary method for related substance profiling
• LC-MS/MS (Liquid Chromatography–Mass Spectrometry) – for trace-level and unknown impurity identification
• Chiral HPLC – for enantiomeric purity testing
• Gas Chromatography (GC) – for residual solvent determination
• ICP-MS – for elemental impurity analysis
• Photostability testing – required due to light sensitivity
Olodaterol is used as a long-acting bronchodilator to help manage COPD, including chronic bronchitis and emphysema, by providing 24-hour symptom control.
It activates β2-adrenergic receptors in the lungs, leading to smooth muscle relaxation, improved airflow, and reduced airway resistance.
• Nasopharyngitis
• Cough
• Dizziness
• Back pain
• Rare: Tachycardia, tremor, or paradoxical bronchospasm
Olodaterol is not approved for monotherapy in asthma. If used, it must be combined with an inhaled corticosteroid (ICS).
Olodaterol is light- and moisture-sensitive, especially in solution. It should be stored in its original sealed, light-protected inhaler at room temperature and used within the labeled period after first activation.
