Pyrimethamine, sold under the brand name Daraprim, is an antiparasitic drug used for the prevention and treatment of toxoplasmosis and malaria. It is a folic acid antagonist that works by inhibiting dihydrofolate reductase (DHFR)—an enzyme essential for DNA synthesis in protozoan parasites.
Pyrimethamine is commonly prescribed:
• Alone or in combination with a sulfonamide (e.g., sulfadiazine) for toxoplasmosis
• Alongside other antimalarials for malaria prophylaxis or treatment in resistant areas
It is available by prescription only, and folinic acid (leucovorin) is often co-administered to counteract bone marrow suppression.
